速冻中低温保护剂浓度对小鼠胚胎发育的影响

摘    要:用快速冷冻方法,冷冻小鼠的2-细胞胚、4-细胞胚、8-细胞胚、桑椹胚和囊胚,低温保护液分别含有0.5,1.5,2.5,4mol/L丙二醇,含10%的犊牛血清和0.25mol/L蔗糖,对于8-细胞以前的小鼠胚胎,快速冷冻保存时丙二醇的浓度在2.5mol/L和4mol/L时效果较好,随着小鼠早期胚胎(囊胚以前)的发育,抗低温冷冻的能力加强。

Abstract:cell,4-cell,8-cell embryos,morula and blastocyst of mice were frozen in differentconcentrations of propylene glycol with rapid freezing method.When the mouse embryos before 8-cell were frozen , it is better to have the glycerol concentrations more than 2.5 mol/L.The em-bryos got higher developing rate frozen with 4 mol/L propyleng glycol.The abilities of enduringlow temperature for mouse embryos were enhanced with embryo development.
