布莱凯特黑牛A-FABP基因表达规律及其肌内脂肪含量研究

摘    要:本实验旨在探究布莱特黑牛脂肪型脂肪酸结合蛋白(A-FABP)基因表达规律,鉴定其与肌内脂肪含量调控的相关性。通过克隆获得A-FABP基因,采用生物信息学方法分析基因结构;利用荧光定量PCR技术研究A-FABP基因在不同组织及不同时期的表达情况;采用PCR-SSCP方法探究基因型与肌内脂肪含量的相关性。结果表明:克隆获得的A-FABP序列与牛、瘤牛、山羊亲缘关系近,同源性高;A-FABP基因在背最长肌中表达量最高,不同时期A-FABP基因的表达量随月龄增加逐渐降低;关联分析显示其多态位点与肌内脂肪含量显著相关(P<0.05),与大理石花纹评分不显著。综上推测,A-FABP基因参与牛肌内脂质代谢相关过程,对调控脂肪的沉积发挥重要作用,可作为肉质性状的辅助标记基因。

Study on the Expression of A-FABP Gene and Its Intramuscular Fat Content in Black Cattle

Abstract:The aim of this experiment was to investigate the expression pattern of Adipocyte fatty acid binding protein(A-FABP) gene in Black cattle, to identify its effects on the regulation of intramuscular fat content. A-FABP gene was obtained by cloning, and the gene structure was analyzed by bioinformatics method. The expression of A-FABP gene in different tissues and different periods was studied by real-time PCR; The A-FABP gene was detected by PCR-SSCP method, and the correlation between genotype and intramuscular fat content was analyzed.The results showed the A-FABP sequence obtained by the clone was close to the bovine, zebu, and goat, and the homology was high. The A-FABP gene has the highest expression in the longissimus dorsi muscle, and A-FABP in different periods. The expression level of the gene gradually decreased with the increase of the age of the month; the correlation analysis showed that the polymorphic locus was significantly correlated with the intramuscular fat content(P<0.05), and the marble pattern score was not significant. In summary, it is speculated that A-FABP gene is involved in the process of lipid metabolism in bovine muscle and plays an important role in regulating fat deposition. It can be used as an auxiliary marker gene for meaty traits.
