芍药茎段愈伤组织诱导的研究

摘    要:以芍药品种‘粉玉奴’茎段为实验材料,1/2 MS培养基为基本培养基,探究不同浓度的6-BA以及不同光质对芍药茎段愈伤组织诱导的影响。试验结果表明,6-BA浓度为0.5mg/L时芍药茎段愈伤组织诱导情况最好;最适宜芍药茎段愈伤诱导的培养基为:1/2MS+Ca^2+(WPM)+PVP 1.0 g/L+NAA 1mg/L+LH 0.5g/L+蔗糖30g/L+琼脂7g/L+6-BA 0.5mg/L;不同的光质条件下,红光更有利于芍药进行愈伤组织诱导。

Abstract:Stem of herbaceous peony(Paeonia lactiflora)‘Fen Yu Nu’was used as explants for callus induction.The effects of different concentration of plant growth regulators 6–BA and light quality on induction were investigated to establish aseptic regeneration system under 1/2MS basic medium.The results showed that the best medium for callus induction was when 6-BA was less than NAA,the best medium to induce callus was 1/2MS+Ca2+(WPM)+PVP1.0 g/L+NAA1.0 mg/L+LH 0.5g/L+sucrose30 g/L+agar7 g/L+6-BA0.5mg/L,and red lihght was more conducive to callus induction under different light quality conditions.
