苯达松敏感致死恢复系的辐射诱变和筛选

摘    要:为了选育对苯达松敏感的水稻恢复系的突变体。利用350 Gy的60Coγ-射线对粳稻恢复系C418和籼稻恢复系浙恢7954进行辐射诱变,M0代舒城育种基地种植并单株收种,收回的M1代分别取6000粒种子于光照培养室中培养,在3叶期用浓度为400.0 mg/L的苯达松溶液涂抹叶片进行筛选。60Coγ-射线诱变对水稻种子有很大的生理损伤,M1代植株育性发生变化的有70%左右;M2代的苗期出现了一定数量的白化苗和黄化苗。用浓度为400.0 mg/L苯达松溶液涂抹叶片筛选出4株C418敏感突变体,突变率为0.07%,筛选出3株浙恢7954敏感突变体,突变率为0.05%。筛选出的突变株基本保持起始亲本的农艺性状。辐射诱变与杂交转育方法相比,具有方法简单、周期短的优点。

Abstract:The study aimed to breeding mutant of rice restorer, which was sensitive to bentazon.Both Japonica restorer C418 and indica restorer Zhehui 7954 were mutated by 350 Gy 60 Coγ-ray irradiation.M0 generation was planted in Shucheng breeding base and the single-plant was harvested for planting.6 000 seeds were chosen from the harvested M1 and cultured in plant growth chamber.The screening was conducted by painting 400.0 mg/L bentazon solution on the leaves in their three-leaf period.60 Coγ-ray irradiation made severe physiological damage to rice seeds and the fertility-changed plants rate was about 70% in M1 generation.The certain amount whitening and etiolated seedlings appeared in the seedling stage of M2 generation.The sensitive mutants of 4 plants of C418 and 3 plants of Zhehui 7954 were screened out by painting leaves with bentazon solution at 400.0 mg/L and their mutation rates were 0.07% and 0.05% resp.The agronomic characters of initial parents were basically kept in the screened mutated plants.Compared with hybrid transferring breeding,the radiation induced-mutation had advantages of simple method and short cycle.
