牛体外受精的精子获能方法对受精率和胚胎发育的影响

从屠宰场获得牛卵巢，采取卵母细胞，成熟培养用ＴＣＭ１９９＋犊牛血清；精子获能处理，以ＢＯ液＋肝素钠为基础，三个试验组分别添加咖啡因、蔡碱和已酮可可碱，受精经分别为８１．３％、８３．３％、８８．９％，均显著高于对照组（Ｐ〈０．０１），三个试验组的囊胚率分别为２０．４％、１９．３％、２１．２％。试验得出以下结论：囊胚率与受经成正比，添加已酮可要碱效果最好。另外组内种公牛成绩也有差异。

Effects of Sperm Capacitation Methods in Cattle In Vitro Fertilization on Fertilization Rate and Embryo Development

Oocytes collected from cattle ovaries at abattoir were cultured for development using TCM199 plus calf serum; the sperm capacitation of control group was treated with BO solution plus sodium heparin; additives including Caffein, Theophylline, and Pentoxifylline were used in the other three experiment groups besides the same treatment of control group. The fertilization rate of three sperm capacitation treatments was 81.3, 83.3, and 88.9% respectively, which was greater (P<0.01) than that of control group, the blastula rate was 20.4, 19.3, and 21.2 respectively. It is concluded that the blastula rate was in proportion to fertilization rate, the Pentoxifylline additive group demonstrated the better result, and the differences of bull performance within group were noted as well.
