安祖花组织培养苗再生体系的优化

摘    要:以安祖花组织培养苗杂AO茎段为试验材料,用正交设计法研究6-BA,2,4-D和KT激素的不同浓度和大量元素中NH4NO3对安祖花愈伤组织诱导的影响;及6-BA,NAA和IBA 3种激素对不定芽分化的影响。结果表明:KT是影响杂AO愈伤诱导的主要因素,不加KT的愈伤诱导率明显高于加KT的愈伤诱导率;各因素影响的主次关系为KT>2,4-D>BA>NH4NO3;最适愈伤诱导的培养基为1/2MS(NH4NO31 100mg/L)+BA(1mg/L)+2,4-D(0.25mg/L);愈伤诱导率为91.2%。6-BA是影响不定芽分化的主要因素,1mg/L 6-BA不定芽诱导率明显高于1.5和2mg/L的诱导率;各因素影响主次关系为6-BA>NAA>IBA;不定芽诱导率的最佳配方为MS+BA(1mg/L)+NAA(0.1mg/L)+IBA(0.1mg/L);不定芽诱导率为83.5%。

Abstract:This paper used miscellaneous AO,stem tissue of Anthurium pedato-radiatum seedlings as the material to study the influence of different concentration of three phytohormone(6-BA,2,4-D and KT)and NH4NO3 on callus induction under the orthogonal design.as well as the influence of three phytohormone(6-BA,NAA and IBA) on adventitious bud differentiation of Anthurium in vitro.The results showed that: the KT is the main factor of callus induction for miscellaneous AO,the callus induction rate in media without KT was higher than the media with KT.The rank of influenced factors was KT>2,4-D>BA>NH4NO3;The optimal media formula for callus induction was 1/2MS + BA 1 mg/L+2,4-D 0.25 mg/L(NH4NO3 1 100 mg/L),the Callus induction rate reached 91.2%.The 6-BA was the main factor of adventitious bud differentiation,the callus induction rate of adventitious bud in the concentration of 1 mg/L 6-BA was higher than in the in the concentration of 1.5 mg/L and 2 mg/L.the rank of three phytohormones was 6-BA>NAA>IBA;the optimal formula for adventitious bud induction was:MS+BA1mg/L+NAA 0.1 mg/L+IBA 0.1 mg/L,the adventitious induction rate reached 83.5%.
