冬凤兰组织培养与快繁技术研究

摘    要:通过探索冬凤兰的组培技术,为冬凤兰的保护和工厂化生产提供技术支撑。以冬凤兰蒴果为原材料,通过不同激素对比,筛选适宜萌芽培养基,并开展了冬凤兰增殖扩繁、生根壮苗、移栽驯化等一系列研究。结果表明:在培养基MS+6-BA 1.0 mg/L+NAA 0.5 mg/L+CM 15%上,萌发时间最早,萌芽率较高;在改良VW培养基上芽分化效果较MS培养基好,芽分化率高,生长势好;在生根培养基VW+IBA 0.5 mg/L+NAA 1.0 mg/L+卡拉胶10 g/L+活性炭0.4 g/L上,生根率达100%;冬凤兰后期需要采用630或650 m L的组培瓶。

Abstract:Tissue culture was conducted to provide technical support to conservation and mass production of Cymbidium dayanum. Capsules of C. dayanum were used as material and cultured for plant regeneration. Different hormones were tried in the MS mediums to select a suitable germination medium. The germinated capsules were then cultured for proliferation, rooting, formation of strong rooted plantlets and transfer.The results showed that the capsule germinated the earliest with the highest germination rate when cultured in the MS+6-BA 1.0 mg/L+NAA 0.5 mg/L+CM 15% medium, that the bud differentiation was higher in the improved VW medium than in the MS, with higher growth vigor, that the rooting rate was 100% when the plantlets were cultured in the rooting medium VW+IBA 0.5 mg/L+NAA 1.0 mg/L+carrageenan10 g/L+0.4 g/L, and that the rooted plantlets were then transferred into tissue culture bottles with the bottle size of 630/650 mL.
