10种南药植物提取物乙酰胆碱酯酶抑制活性的筛选模型研究

摘    要:采用筛选模型对10种南药样品乙酸乙酯提取物的乙酰胆碱酯酶抑制活性进行了评价。结果表明:10种南药(益智、鸡血藤、决明子、巴戟天、溪黄草、金樱子、化橘红、佛手、绞股蓝、五指毛桃)均具有不同程度的乙酰胆碱酯酶抑制活性。当提取物浓度为1.0 mg/mL时,溪黄草、益智、化橘红、绞股蓝、五指毛桃表现出较强的抑制活性,平均抑制率大于50%,分别为89.59%,77.10%,67.54%,52.62%,52.29%,其他5种南药的平均抑制率在20.81%~49.81%之间;提取物浓度为0.1 mg/mL时,溪黄草、益智、佛手仍表现出相对较强的抑制活性,平均抑制率大于25%,其他7种南药的平均抑制率在6.07%~24.78%之间。应该深入研究复方中药治疗老年痴呆的药效物质基础,加大对南药资源的开发利用。

Evaluation of Acetylcholinesterase Inhibitory Activity of 10 Southern Chinese Herbs by Screening Model

Abstract:In the present study, AChE activity of ethyl acetate-soluble fractions from 10 southern Chinese herb samples were screened by a screening model. The results indicated that all these 10 samples exhibited AChE inhibitory activities. At 1.0 mg/mL of extracts concentrations, higher inhibitory activity appeared in Rabdosia serra ( Maxim.) Hara, Alpinia oxyphylla, Citrus grandis cv. Tomentosa, Gynostemma pentaphyllum, and Radix Fici Hirtae, with inhibition ratios of 89.59%, 77.10%, 67.54%, 52.62%, and 52.29% (all of the above more than 50%) respectively, and the inhibition ratios of the other 5 samples at this concentration were between 20.81%and 49.81%. At 0.1 mg/mL of extracts concentrations, Rabdosia serra (Maxim.) Hara, Alpinia oxyphylla, Citrus medica L. and var. sarcodactylis Swingle demonstrated higher inhibitory activites, with average inhibition ratios more than 25%, while the inhibition ratios of the other 7 samples at this concentration were between 6.07% and 24.78%. Exploring of southern Chinese herbs should be strengthened in searching components for treatment of Alzheimer's Disease.
