摘    要:以超甜玉米自交系S1、S2、S3和粤甜3号等为材料,建立了超甜玉米高效再生系统。研究结果显示,超甜玉米基因型、幼胚的大小是影响胚性愈伤组织生成的主要因素,幼胚长度在0.5-1.2mm之间时,胚性愈伤组织的诱导频率最高。培养基中蔗糖浓度和细胞分裂素浓度对胚性愈伤组织的分化和小植株再生频率有重要影响,低浓度的蔗糖(20g/L)和6-BA(2mg/L)获得的再生频率较高。继代培养中ABA处理可使愈伤组织的再生频率提高1.68倍,IBA浓度在3.0mg/L,可以明显提高小植株生根。

Abstract:Using super sweet corn inbreeding lines S1, S2, S3 and hybrid Yue Tian No.3 as materials, the efficient regeneration systems were established. The research showed that the genotype and length of immature embryo of super sweet corn were the key factors influencing the embryogenic callus development, 0.5-1.2mm immature embryo were better for embryogenic callus inducing. contents of sugar and cytokinins in medium were the main factors for plantlet regeneration, low content of sugar( 20g/L) and 6-BA(2mg/L)or Zeatin (3 mg/L) are more fit to the regeneration, and the rate of regeneration could be increased 1.68 times when callus treated with ABA in sub-culture for 3 weeks. IBA could improve plantlet rooting, and the content of 3.0mg/L was the best.
