转化型人参皂苷抗MDV及诱导肿瘤细胞凋亡的作用初探

摘    要:以转化型人参皂苷的8个组,按12.5μg·mL-1的剂量,在体外鸡胚成纤维细胞上进行抗MDV-RB1B强毒株空斑减数试验和诱导肝癌7402细胞株、大鼠脑胶质瘤细胞C6株细胞凋亡试验,用光学显微镜观察、琼脂糖凝胶电泳检测分析DNA损伤。结果表明:转化型人参皂苷6、7号组,在感染阻断、增殖抑制、直接杀灭3种方式的抗MDV空斑减数率都达到阿昔洛韦(ACV)抗MDV空斑减数率的80%,转化型人参皂苷4、6号组对MDV-RB1B株的直接杀灭作用与ACV的杀灭作用相同;转化型人参皂苷8号组诱导肝癌7402株细胞48h后,有60%的瘤细胞凋亡,对大鼠脑胶质瘤C6株细胞也有诱导凋亡的作用。

Abstract:The role of eight transformed ginsenosides (TG) against MDV-RB1B and inducing apoptosis in hepatocellular carcinoma cell line 7402 cells and rat glioma cell line C_6 cells by plaque reduction assay in vitro were investigated. The DNA damage was determined by microscopy and agarose gel electrophoresis. The results showed that the infection and reproduction of MDV in chicken embryo fibroblast (CEF) could be blocked or inhibited by TG-6 and TG-7, MDV could also be inactivated directly by TG-6 and TG-7. The plaques of MDV in CEF were reduced up to 80% compared to those of ACV group. The inactivating role of TG-4 and TG-6 to MDV-RB1B cells was equal to that of ACV group. 60% apoptosis cells for hepatocellular carcinoma cell line 7402 cells were found when the cells were exposed to TG-8 for 48 h and induction of apoptosis was also found in rat glioma cell line C_6 cells.
