黄花乌头种子萌发与水溶性内源抑制物质研究

摘    要:[目的]为提高黄花乌头种子发芽率提供理论依据。[方法]以黄花乌头种子为材料,研究不同浓度赤霉素对种子发芽率的影响,测定该种子粗提物对白菜种子萌发及幼根的影响以及温水和溶剂浸种去除内源抑制物质的效果。[结果]用200μg/ml赤霉素处理时,黄花乌头种子发芽率和发芽势最高,为79.6%和52.4%,比对照高40.9%和71.4%。室温0~4℃变温浸种处理的种子发芽势和发芽率最高;黄花乌头种子的粗提物有较强的抑制活性,当粗提物浓度为0.28 g/ml时,白菜种子的相对发芽率为20.2%,幼根相对长为11.2%。黄花乌头种子的相对发芽率为15.2%,相对发芽势为6.4%;温水浸种可除去黄花乌头种子中大量内源抑制物质。乙醇浸种时浸泡液的抑制活性最强。[结论]200μg/ml赤霉素变温浸种处理,可提高黄花乌头种子的发芽率。

Abstract:[Objective] The study aimed to provide the theoretical basis for increasing the seed germination rate of Aconitum coreanum.[Method] With A.coreanum seeds as material,the effects of GA3 with different concn.on seed germination rate were studied,and the effect of the crude extracts from the seeds on the germination rate of cabbage seed and its radicle growth and the effects of seed soaking with warm water and solvents on removing inner inhibitory substances were detected.[Result] The germination rate and germinating energy of A.coreanum seeds were the highest when the seeds were treated by 200 μg/ml GA3,being 79.6% and 52.4%,and were 40.9% and 71.4% higher than that of CK resp.The seed germinating energy and seed germination rate were the highest in the treatment of seed soaking at variable room temperature(0-4 ℃).The crude extracts from A.coreanum seed had stronger inhibitory activity.When the crude extracts was 0.28 g/ml,the relative germination rate of cabbage seed was 20.2% and the relative length of radicle was 11.2%.The relative germination rate of A.coreanum seed was 15.2%,and the relative germinating energy was 6.4%.Massive inner inhibitory substances in A.coreanum seeds could be removed by seed soaking with warm water.The inhibitory activity of soaking solution was strongest when the seeds were soaked by alcohol.[Conclusion] The seed germination rate of A.coreanum could be increased by seed soaking treatment with 200 μg/ml GA3 at variable temperature.
