马铃薯病毒试管苗保存技术及病毒侵染力的研究

摘    要:无菌条件下切取毒源马铃薯茎尖于MS培养基中培养,当毒源试管苗长至10 cm时切取单节段扩繁。通过免疫电镜法、电镜负染法和DAS-ELISA法鉴定,筛选出4管纯毒源材料。筛选出的试管苗毒源经18个月保存病毒仍存在,将毒源试管苗移至花盆中栽培,试验管苗表现出马铃薯病毒A(PVA)侵染症状,用毒源马铃薯汁液接种的指示植物也表现典型PVA症状,说明毒源试管苗保存的PVA具有侵染力。用试管苗保存马铃薯病毒,可提高毒源保存效果。

Abstract:The test was done like this: cutting the stem tips of potato plantlet in the state of asepsis and cultivating them in the tube with MS,further culturing them by cutting them into single sections when the stem tips grew to 10cm.Selecting 4 tubes of test-tube plantlets through immunity electronic microscope capturing,electronic microscope negative staining and DAS-ELISA.The selected drug sources still existed after 18 months virus preservation and performed as infected by PVA when the drug source test-tube plantlet was planted into the flowerpots.Indicator plants showed the typical symptom of PVA after inoculating with the juice of drug source potatoes.The result indicates that PVA in test-tube plantlet has infectious potential.Preserving PVA in potato test-tube could improve the preservation effects.
