摘    要:采用两步稀释法 ,在绵羊冷冻精液中添加维生素E ,精液稀释平衡后精子活率 ,试验组 (0 .6 35 )高于对照组(0 .5 75 ) ,但无显著性差异 (P >0 .0 5 ) ;而解冻后 ,试验组活率 (0 .4 35 )极显著高于对照组 (0 .36 5 ) (P <0 .0 1) ;精子顶体总异常率 ,试验组 (33.72 % )极显著低于对照组 (4 4 .35 % ) (P <0 .0 1) ,其中试验组顶体膨胀率和脱落率与对照组分别为 1.2 8% ,2 3.0 8%与 2 .6 8% ,2 8.96 %。添加维生素E可以降低冷冻对精子顶体的损害程度 ,提高精液品质。试验也证明在解冻后保存 1h内 ,精子活率呈现缓慢下降的趋势 ,但 1h后活率急剧下降 ,因而 ,绵羊冻精颗粒解冻后应在 1h内输精完毕

Abstract:The objective of this study was to determine the effect of supplementation of vitamin E (VE) in diluent on the ram semen quality of frozen-thawed semen by 2-step method of cryopreservation procedure. The results indicated that the motility of balanced semen in treatment group (0.635±0.023) was increased compared to the control (0.575±0.028), but not significantly (P>0.05); However, the motility of the frozen-thawed semen in treatment group (0.435±0.020) was significantly (P<0.01) higher than that of the control (0.365±0.026). The percentage of membrane integrity disrupted of frozen-thawed seminal acrosome was reduced significantly by supplementation of VE (P<0.01), 33.72% in treatment group vs. 44.35% in the control respectively. Both the percentage of the swelled and desquamated acrosome were decreased (1.28%±0.58%, 23.08%±1.45%) in the treatment group than that of the control (2.68%± 0.59% , 28.96%±3.14%).It indicated that the motility of frozen-thawed semen was declining tardily within one hour after thawed, however it was sharply decreasing beyond that limit of one hour. So the artificial insemination of frozen-thawed semen should be carried out within one hour after thawed.
