黄粉虫幼虫抗菌肽的诱导特性及电泳组分研究初报

摘    要:对黄粉虫幼虫分别进行空白对照、菌刺、菌饲、热激和超声波四种处理,并于处理后1d、2d、3d、4d、5d分别提取黄粉虫幼虫体内的抗菌肽,做抗大肠杆菌(革兰氏阴性菌)和抗金黄色葡萄球菌(革兰氏阳性菌)试验及交叉抑菌试验,测定其抑菌圈大小,确定其抗菌活性和诱导特异性,同时还进行抗菌肽热稳定性及电泳试验。结果表明:水浴后提取的抗菌液抑菌效果明显,四种处理的黄粉虫幼虫均能产生抗大肠杆菌/金黄色葡萄球菌物质。其中抗金黄色葡萄球菌效果要略好于抗大肠杆菌效果,其抗菌效果显著性依次为:菌饲、对照、热激和超声波、菌刺,而抗大肠杆菌效果显著性则依次为:菌饲、菌刺、热激和超声波、对照。交叉抑菌试验表明黄粉虫幼虫体内抗菌肽的产生不具有诱导特异性。电泳图谱显示黄粉虫抗菌肽含有5种肽组分。

Abstract:The larva of Tenebrio molitor L. were treated with contrast, pricking with bacteria, feeding with bacteria, heat shock and uhrasonication in each experiments respectively. In 1 d ,2d ,3d ,4d ,5d, antibacterial peptide were abstracted from the larva of Tenebrio molitor L. The experiments of the anti - E. coli ( G - ) and the anti - Staphylococcus aureus( G ^+ ) and cross - anti - bacterial were conducted. The anti - bacteria activity and inducement characteristics of anti - bacterial peptide from the larva of Tenebrio molitor L. were distinguished when the size of bacteriostasis ring was measured. At the same time, comparative experiments between 5 min boiled water bath in 100℃ and without were done to make sure it has thermal stability. Electrophoresis test was done, too. The results showed that the antibacterial substance which was treated in boiled water bath had significantly bacteriostatic effect. The larva of Tenebrio molitor L. which were treated by the 4 ways above all produced antibacterial material which had bacteriostatic effect on E - coli and Staphylococcus aureus. The bacteriostatic effect on Staphylococcus aureus was better than that on E - coil The significance of anti - Staphylococcus aureus experiments in each treatment were sized down: feeding with bacteria, heat shock and uhrasonication, contrast , pricking with bacteria, in turn. And the significance of anti - E. coli experiments in each treatment were sized down: feeding with bacteria, pricking with bacteria, heat shock and ultrasonication, contrast. The experiments of cross - anti - bacterial showed that producing of antibacterial peptide abstracted from the larva of Tenebrio molitor L had non - specific induction. Electrophoresis test indi- cated that the anti - bacterial peptide from the larva of Tenebrio molitor L.. were composed of 5 components.
