利用ISSR对矮牵牛品种遗传多样性的聚类分析

摘    要:为了揭示矮牵牛品种资源的遗传多样性,并更好地应用于育种工作,利用ISSR分子标记技术对142份矮牵牛材料进行了遗传多样性研究。在供试材料中,筛选到具有多态性的ISSR引物17个,共扩增出多态性条带132条,平均每个引物扩增的多态性条带数为7.8条,多态性条带比率为82.5%。材料间遗传相似系数范围在0.401 5~0.931 8间,这说明矮牵牛具有丰富的遗传多样性。通过软件计算结果表明,142个矮牵牛品种平均有效等位基因数为1.653 5±0.285 3,Nei基因多样性指数平均为0.374 4±0.123 6,平均Shannon信息多样性指数为0.552 6±0.148 6。将矮牵牛材料聚类结果与其花色、花型、植株类型、品种来源作比较,呈现一定的相关性,这为矮牵牛优良品种的选育奠定了一定的理论基础。

Abstract:In order to reveal the genetic diversity and better apply for breeding,Inter-Simple Sequence Repeat(ISSR) molecular markers were used to detect the genetic diversity in 142 varieties of Petunia hybrida Vilm.Seventeen ISSR primers were selected for ISSR-PCR.A total of 132 polymorphic bands were amplified,and the average polymorphic bands of every primer was 7.8 with 82.5% of polymorphism-strip ratio.The genetic similarity coefficient between material ranged from 0.401 5 to 0.931 8,which showed the P.hybrida has a wealth of genetic diversity.The software calculation results showed that 142 P.hybrida varieties average effective numbers of alleles,the average index of Nei genes diversity,and the average index of Shannon information diversity were 1.653 5 ± 0.285 3,0.374 4 ± 0.123 6,0.552 6 ± 0.148 6,respectively.There was certain correlative relationship between the clustering results and the plant characters including color,pattern,type and varieties comparison of source,which provided molecular theoretical basis for the breeding of P.hybrida.
