农杆菌介导的大豆胚尖遗传转化体系中基因型的选择及抑菌条件的优化

摘    要:大豆基因型对农杆菌介导大豆胚尖遗传转化体系的转化效率有很大影响,同时有效地抑制农杆菌是该体系的重要环节。研究选择3个东北地区主栽的大豆品种,以胚尖为外植体进行农杆菌介导的遗传转化,比较3个大豆品种的诱导率、伸长率和生根率,并在芽诱导、伸长及生根阶段采用不同的抗生素浓度配比,优化抑菌条件。实验结果表明3个品种中,黑农37为最佳受体品种;抗生素浓度对大豆胚尖遗传转化有很大影响。在芽诱导和伸长阶段添加头孢霉素300mg/L+羧苄青霉素150mg/L抑菌效果较好;而在生根过程中,不添加抗生素为最佳条件。

Abstract:Genotype is one of the factors affecting the efficient of soybean transformation,and effective bacteriostasis is very important point in the Agrobacterium-mediated transformation.Overuse of antibiotic will cause the suppression in induction,elongation and rooting.We used the embryonic tips of Heinong 37,Hefeng 35 and Dongnong 42 as explants and obtained the rate of induction,elongation and rooting,which were treated with different ratio and concentration of cephalosporin(Cef) and carbenicillin(Cb) combination.The results indicated that Heinong 37 was the best receptor and 300 mg/L of Cef + 150 mg/L of Cb had better bacteriostatic effects in the stage of bud induction and growth,but it’s optimal without antibiotic in rooting period.
