甘薯线虫病品种抗性的PCR检测

摘    要:根据甜菜抗线虫病基因序列,人工合成了三条上游引物和三条下游引物,从中筛选出一对引物,对11份经抗线虫病常规鉴定的甘薯品种(包括6份高抗,2份抗病,3份感病)进行了PCR检测.结果表明:6份高抗品种和1份抗病品种出现了一条630 bp左右大小的特异片段,而3份感病品种未出现任何条带.这与常规鉴定的结果基本吻合,证明分子检测的可行

Abstract:According to the sequence of nematode resistant gene in sugar beet, three forward primers and three reverse primers were produced by synthetic. A pair of primers were selected among them. 11 sweet potato cultivars (including 6 nematode high resistant, 2 resistant and 3 sensitive cultivars by conventional identification)were detected by PCR. The results showed that a special fragment of about 630 bp appeared in 6 high resistant and 1 resistant cultivars, and nothing in 3 infected cultivars. The result was in correspondence with that of conventional identification, and suggested the feasibility of PCR detection.
