苎麻花叶病病原的研究

摘    要:本研究结果表明,用高灵敏度的聚丙烯酰胺凝胶双向电泳银染法,在感病植株叶片中检测不到环状小分子RNA;用常规方法对患病植株的根、茎、叶进行分离培养,未发现细菌或真菌,因此,基本上排除了类病毒、细菌、真菌作为苎麻花叶病病原的可能性,用植物病毒提纯方法,在感染植株叶片分离物中发现一种长约1000nm,宽约11 nm的线状病毒,而在健康植株叶片中,未发现这种病毒粒子,这种病毒粒子经超离心浓缩后,可感染指示植物苋色藜,表现感病症状,在感病苎麻叶片的超薄切片中,可观察到大量内含体,该病毒是否即为苎麻花叶病的唯一病原,尚需进一步探讨。

Abstract:Low molecular weight circular RNA can not be detected in the infectedramie leaves with Two-Directional PAGE-silver Staining. Incubating the root,stem, and leaves of infected plant finds no bacteria or fungi. So the possibi-lity that the disease is caused by viroid, bacteria, and fungi can be ruled out.Through extraction and purification of plant virus, a thread-like virus about1000 nm long and 11 nm wide is found in the infected leaves, but not in thehealthy leaves. The virus can infect an indicat plant Cohempodium amaranti-color Coste & Revn. and cause local necrosis on its leaves. The inclusionbodies are observed in the ultrathin section of infected ramie cell. Furtherstudy is needed to decide whethe the virus is the only agent of the disease.
