基于mt DNA COI基因的宁夏烟粉虱及温室白粉虱分子鉴定

摘    要:基于线粒体COI基因(Cytochrome coxidase subunit I gene)对宁夏地区粉虱进行分子鉴定。选取宁夏19个粉虱地理种群的77个样本作为试验材料,用Bioedit 7.2.5软件对COI基因序列进行读取、整合、对齐比对处理,用软件MEGA 7.0中的Kimura 2-parameter模型计算种内与种间遗传距离,并用邻接法(NJ法)构建系统发育树,用软件DNASP 5.10进行遗传多样性的分析,用软件Network 5.0构建单倍型网络图。结果显示,77个样本中的46个为Q型烟粉虱(Bemisia tabaci),31个为温室白粉虱(Trialeurodes vaporariorum),表明目前Q型烟粉虱已经传入宁夏银川市、石嘴山市、中卫市等,亟需对其危害和扩散进行有效控制。

Abstract:The purpose of this study is to carry out the molecular identification of whiteflies in Ningxia based on mitochondrial COI gene (cytochrome coxidase subunit I genes). The COI gene sequences of 77 individuals from 19 geographical populations of whiteflies in Ningxia were amplified. The COI gene sequences were edited and aligned using Bioedit 7.2.5 software. The intra-and inter-species genetic distances were calculated with MEGA (Version 7.0) using the Kimura-2-Parameter model. The neighbor-joining (NJ) trees were reconstructed using MEGA (Version 7.0) software. Genetic diversity was analyzed by DNASP 5.10 and haplotype Network graph was constructed by software Network 5.0. The results showed that 46 of the 77 individuals were Q biotype of Bemisia tabaci and 31 were Trialeurodes vaporariorum. It was shown that Q biotype of B.tabaci had been introduced into Ningxia, including Yinchuan, Shizuishan, Zhongwei city and so on, so it is urgent to control its harm and diffusion effectively.
