平菇原生质体制备及再生培养研究

平菇菌丝原生质体的释放以菌龄４～６ｄ的幼嫩菌丝，用２％Ｌｙｗａｌｚｙｍｅ采用０．６ＭＫＣｌ为渗稳剂在ｐＨ值为５．５、温度为３５℃、酶解２～２．５ｈ生长较好，可达１０７个／ｍＬ，孢子释放原生质体在１．５％溶壁酶和１．５％纤维素酶混合作用，同样外界条件下原生质体转化率可达１００％。原生质体再生以２号培养基在０．６Ｍ甘露醇的作用下，用载片悬浮滴式再生法，再生率可达１２．３％。

The more protoplasts of pleurotus mycelium are released from the mycelia at age of 4 to 6 days under the conditions as PH5.5, 35℃,2～2.5h enzyme action with 2% lywallzyme and 0.6 mol/L KCl as osmotic stabitizor, the number is 10 7/mL. Under the same external conditions, production efficiency of protoplast can reach 100% while the spore is put in the mixture of 1.5% lywallzyme and 1.5% cellocidin enzyme. Regeneration ratio of protoplast cultivated in the No.2 medium with 0.6 mol/L mannitol as osmotic by hanging drop method on concave can reach 12.3%.
