摘 要: | 平菇菌丝原生质体的释放以菌龄4~6d的幼嫩菌丝,用2%Lywalzyme采用0.6MKCl为渗稳剂在pH值为5.5、温度为35℃、酶解2~2.5h生长较好,可达107个/mL,孢子释放原生质体在1.5%溶壁酶和1.5%纤维素酶混合作用,同样外界条件下原生质体转化率可达100%。原生质体再生以2号培养基在0.6M甘露醇的作用下,用载片悬浮滴式再生法,再生率可达12.3%。

Abstract: | The more protoplasts of pleurotus mycelium are released from the mycelia at age of 4 to 6 days under the conditions as PH5.5, 35℃,2~2.5h enzyme action with 2% lywallzyme and 0.6 mol/L KCl as osmotic stabitizor, the number is 10 7/mL. Under the same external conditions, production efficiency of protoplast can reach 100% while the spore is put in the mixture of 1.5% lywallzyme and 1.5% cellocidin enzyme. Regeneration ratio of protoplast cultivated in the No.2 medium with 0.6 mol/L mannitol as osmotic by hanging drop method on concave can reach 12.3%. |
