摘    要:选用石斛兰茎尖和根尖为外植体,以不同的激素浓度组合的MS培养基,研究植物激素对石斛兰组织培养中原球茎的诱导、增殖、生根的影响.结果表明,茎尖能够诱导出原球茎,而根尖几乎不能.最适宜的石斛兰原球茎诱导培养基是MS+6-BA 0.5mg/L+NAA 2.0 mg/L,其原球茎出愈率达85%;适宜原球茎分化的培养基是Ms+6-BA 4 mg/L+NAA 1 mgL,其分化率达90%;最适宜石斛兰诱导根的培养基应为1/2MS+2.0~3.0 mg/L IBA,生根率达100%.

Abstract:The effects of different hormone levels on the inducement,proliferation and radieation of protocorm in tissue culture of dendrobium were studied and the stem apex and root tip of dendrobium were taken as explants.The results showed that the protocorm could be induced from stem apex,but barely from root tip.The optimum inducement medium for protocorm of dendrobium was MS+6-BA 0.5 mg/L+NAA 2.0 mg/L,which inducing rate reached 85%;the optimum differentiation medium for protocorm of dendrobium was MS+6-BA 4 mg/L+NAA 1 mg/L,which differentiation rate reached 90%;the optimum medium for induced root of dendrobium was 1/2 MS+2.0~3.0 mg/L IBA,which rooting rate reached 100%.
