半夏不同变异类型的RAPD分析

摘    要:为了利用RAPD技术研究半夏的各种变异类型。以栽培多年的6个不同变异类型的半夏为实验材料,取每一变异类型中的5~10株新鲜叶片,提取总基因组DNA。从100条随机引物中筛选出17条重复性好、条带清晰、能体现半夏性状变异类型差异的引物进行RAPD扩增。RAPD分析结果表明,17个RAPD引物扩增出了81条带,其中55条为多态带,占67.9%,表明半夏不同变异类型间存在分子水平的遗传差异。该研究为半夏的品种改良与栽培奠定了基础。

Abstract:The study was constructed to make use of RAPD technology to study the variance types of Pinellia ternata(Thunb.) Breit.With 6 different variance types of P.ternata cultured for many years as experiment materials,total genomic DNA was extracted from fresh leaves of 5~10 plants for each variance type.17 primers that could embody the differences in various variance-types with good repeatability and clear bands were screened from 100 random primers for the RAPD amplification.RAPD analysis showed that 81 bands were produced by the amplification of 17 RAPD primers,with 55 bands being polymorphic(being 67.9%),which indicated that there were genetic difference in the molecular level among different variance types.The study laid the foundation for variety improvement and cultivation of P.ternata.
