摘    要:以番茄为材料,通过对扩增长度多态性(AFLP)反应体系中的几个关键参数进行优化,建立了适合于番茄的AFLP分子标记体系。结果表明,用于反应的DNA采用CTAB法提取较好,酶切的基因组DNA以100 ng为宜,酶切-连接采用一步法,37℃条件下进行,反应时间为10 h,预扩增产物的稀释倍数以30倍为宜,采用优化好的体系,29对引物组合在两份供试材料均可扩增出稳定清晰的带纹60~100条。本研究为番茄的分子标记克隆及抗病育种的辅助选择提供了有力工具。

Abstract:AFLP(Amplified fragment length polymorphism) analysis system was established with tomato by optimizing some factors of the AFLP technological system.The results showed that the dosage of template DNA should be 100 ng,the time of digestion and ligation should be 10 h,and the products of preamplification should be diluted to 30 times at 37 ℃.With the optimized system,the AFLP fingerprinting of tomato materials amplified by primer combination 29 pairs have been obtained,which had 60-100 clear bands.Our results provided an useful tool to study the molecular cloning of tomato and were helpful for molecular assisted selection of tomato resistant breeding.
