伪鹅观草高分子量麦谷蛋白基因启动子的克隆

摘    要:通过PCR克隆的方法,从二倍体伪鹅观草St基因组中克隆到了高分子量麦谷蛋白亚基基因Glu-1St1的启动子,该序列全长959bp,序列比对及结构分析表明,它与麦类高分子量麦谷蛋白亚基基因的启动子序列的一致性很高,达到82%,包含有典型麦谷蛋白亚基基因启动子的顺式作用元件。系统进化分析表明,它与大麦的D-hordein基因启动子的进化关系比较近,属于典型的x型亚基基因的启动子。

Abstract:The promoter sequence with 959 bp of Glu-1St1 was obtained by genomic PCR experiment in diploid Pseudoroegneria stipifolia which contained St genome.The structural analysis indicated that the sequence had very high identity with the promoters of HMW-GS(high molecular weight glutenin subunits) genes in Triticum,and including an identical set of cis-elements found in promoter regions of previously characterized wheat HMW-GS genes.Phylogenetic analysis indicated that the promoter of Glu-1St1 was closely related with that of D-hordein,and clustered to x-type subunit gene promoters.
