番茄果实酸性转化酶基因cDNA片段的克隆

摘    要:根据酸性转化酶基因的保守序列设计引物 ,采用RT PCR方法 ,从番茄果实总RNA中扩增出目标cDNA片段 ,克隆到pMD18 T载体中。在所测的 10 38个核苷酸中 ,与GenBank中登记号为M810 81的番茄果实酸性转化酶基因高度同源 ,同源性为 99%。该基因片段在GenBank中已登记 ,登记号为AF4 90 5 30。

Abstract:PCR primers were designed based on the consensus domain of some acid invertase genes in GenBank. A cDNA fragment was amplified from the total RNA isolated from toamto fruit via RT PCR and cloned into pMD18 T vector. The determined sequence showed that the amino acid sequence is 99% homologous to that of the corresponding region of acid invertase gene (Access No. M81081) in GenBank. This confirmed that we cloned a cDNA fragment from tomato fruit successfully and its access number is confirmed as AF490530 in GeneBank.
