玉米大斑病抗性基因Ht2定位区域内候选序列的生物信息学分析

摘    要:利用计算机技术对与玉米大斑病抗性基因Ht2紧密连锁的分子标记umc89和umc48a定位区域内全部序列进行生物信息学分析,共发现101条ORF序列,其中16条ORF序列可编码蛋白质结构域。将这16条序列与已克隆的23条抗病基因核苷酸序列进行BLAST比对及进化树构建,并分析其与已知抗病基因的相似性及亲缘关系。结果发现,序列pco115381g和pco110559-1与编码水稻白叶枯病基因Xa1相似性高、亲缘关系近,而且具有植物抗性基因的高度保守结构域特征,可能与玉米大斑病抗性基因Ht2有关。

Abstract:All candidate sequences linked with Ht2(molecular markers from umc89 to umc48a) were analyzed by reference computer technology in this research. There were 101 ORF sequences were found in this area, of which 16 candidate sequences had the function of encoding protein domain. The 16 candidate sequences were used to blast with the cloned R-gene and establish phylogenetic tree. Sequence analysis indicated that candidate sequences pco115381g and pco110559-1 had characteristics of relatively high identity and homology to Xa1(resistant to bacterial blight). In addition, there had another characteristic of high conserved NBS domain. These results revealed that the candidate sequences pco115381g and pco110559-1 were related to Ht2.
