影响彩色马蹄莲瓶内结球因素研究

摘    要:彩色马蹄莲瓶内结球试验结果表明:提高糖浓度可以促进瓶内结球,糖浓度为100g/L时瓶内结球率和生长效果最好,添加多效唑的效果不及提高糖浓度,基质培养基在糖或多效唑浓度较低时结球效果好,而固体培养基在糖和多效唑浓度分别高于100g/L和15mg/L时结球效果更好;光照条件有利于小籽球的形成和生长。在MS+NAA0.5mg/L+糖100g/L+多效唑15mg/L的固体培养基中结球效果最佳,结球率达到73.8%,大于5mm的籽球比例达到48.1%,每瓶球重达到0.55g。

Abstract:The effects of sucrose,PP333,culture medium and illumination on the formation of micro-tuber from isolated calla lily were investigated.The results showed that high concentration of sucrose could promote the formation of micro-tuber,and the best concentration was 100 g/L.The effect of sucrose on micro-tuber formation was better than that of PP333.Low concentration of sugar or PP333 in the medium with perlite and turf could promote micro-tuber formation.Moreover,the solid medium with above 100 g/L sugar and more than 15 mg/L PP333 had better effect on micro-tuber formation.Illumination could promote micro-tuber formation and growth.The best solid culture medium for the formation of micro-tuber was MS NAA 0.5 mg/L sugar 100 g/L PP333 15 mg/L;on the above best medium,the formation percentage of micro-tuber was 73.8%,the percentage of micro-tuber larger than 5 mm was 48.1%,the weight of micro-tuber per bottle was 0.55 g.
