粗提牛白细胞抗菌肽对小鼠免疫功能的影响

摘    要:将粗提牛白细胞抗菌肽分别以低、中、高剂量(1.5,7.5,15 g·L~(-1))灌胃小鼠,0.2 mL·只~(-1);对照组分别为灌施体积分数为0.01%醋酸溶液和10 g·L~(-1)的牛血清白蛋白各0.2 mL.测定小鼠的细胞吞噬功能和免疫器官指数;同时采用流式细胞术测定小鼠外同血中T淋巴细胞亚群的总数及各亚群的百分数.结果表明,7.5 g·L~(-1)的粗提牛白细胞抗菌肽能明显提高小鼠的免疫器官指数(P<0.05),增强小鼠的细胞吞噬功能(P<0.05)以及小鼠外周血CD3~+,CD4~+及CD8~+T淋巴细胞的百分数(P<0.01),而高剂量组比中剂量有所降低.7.5 g·L~(-1)的粗提牛白细胞抗菌肽能明显增强小鼠的免疫功能,提高机体的免疫水平.

Abstract:Bovine blood was collected and the antimicrobial components were extracted from leukocytes through ammonium chloride lyses, sonication, centrifugation and freeze-drying. The extract was admin-istrated for two weeks by means of gavage in a daily dose of 0. 2 mL at a protein concentration of 1. 5, 7.5, 15 g·L~(-1) respectively, and the control groups were separately fed with 0. 2 mL of 0. 01% acetic acid and 10 g·L~(-1) BSA . The serum phagocytic rate, phagocytic index, and weight of the immune or-gans ( spleen and thymus) were measured, whilst the total number and subgroups of T lymphocytes were mounted through flow cytometry ( FCM). The results shows that at a dosage of 7. 5 mg protein content, the extract improves the serum phagocytic index, which indicates the ability of phagocytosis is enhanced (P<0.05) ; the immune organ weight index increases significantly (P < 0. 05) ; the per-centages of CD3~+ , CD4~+ , and CD8~+ T cells are also greatly improved ( P < 0, 01) . It may be conclu-ded , therefore, that the bovine leukocyte extract can improve the level of both innate immunity and a-daptive immune responses so that it can be promisingly applied in both human medicine and veterinary fields.
