白木香rDNA ITS序列测序鉴定的初步研究

摘    要:以改良的CTAB法提取白木香的基因组DNA,运用通用引物对其rDNA的ITS区进行PCR扩增,并对扩增产物分别进行直接测序和克隆测序。结果表明,白木香rDNA的ITS区序列存在稳定而又明显的差异,白木香rDNA序列差异可能主要集中在ITS1区,存在5处变异位点,ITS2区存在1处变异位点。

Abstract:The genomic DNA of Aquilaria sinensis(Lour.) Gilg was extracted from yong leaves by improved CTAB method,the ITS sequence was amplified by PCR with universal primers of ITS and PCR products were direct sequenced and clone sequenced.The results showed that there was a clear difference in rDNA ITS of Aquilaria sinensis(Lour.) Gilg,and there were five variable sites in the ITS1 area,a variation of sites in the ITS2 area.
