南阳牛小腔卵泡卵母细胞体外成熟的研究

摘    要:探讨了不同采集方法、牛卵泡液(bFF)添加浓度、培养时间和不同培养体系对南阳牛小腔卵泡卵母细胞体外成熟的影响。结果表明:组织破碎法、剥离法和切割法平均每个卵巢采到的卵母细胞数三者之间无显著差异,但用切割法所得的卵母细胞经体外培养,其成熟率显著高于剥离法和组织破碎法;成熟液中添加10%bFF组的卵母细胞成熟率显著高于对照组和添加5%bFF组,但其卵母细胞成熟率与20%bFF组差异不显著;在不同培养时间、不同培养体系试验设计中,以30~32h组、TCM199+OCS+FSH+LH+LIF组的卵母细胞成熟率较高。

Abstract:This experiment studied the effects of ova pick-up methods, bFF,culture period and different culture systems on in vitro maturation of Nanyang cattle oocytes derived from small antral follicles.Results showed that: the number of oocytes picked by tissue-breaking method was the most(12.04),that picked by cutting method was the least(8.63),but there were no significant difference among these three methods(P>0.05);the maturation rate of oocytes from cutting method(21.26%)was higher significantly than that from peel-off method(15.21%) and tissue-breaking method(13.15%)(P<0.05);supplementation of 10% bFF in maturation medium significantly improved the maturation rate of oocytes(16.67%),higher than that of the ck(9.09%)and that of supplementation of 5% bFF(11.76%)(P<0.05);the maturation rates of culture period 30-32h(12.50%)and culture system TCM199 OCS FSH LH LIF(21.21%)were higher than those from the corresponding treaments.
