黄瓜内生枯草芽孢杆菌B504发酵配方优化研究

摘    要:利用SAS软件对黄瓜内生枯草芽孢杆菌(Bacillus subtilis)B504发酵培养基进行优化。首先用Plackett-Burman法筛选出3个重要因素,通过最陡爬坡试验逼近最佳响应面区域,最后采用Box-Behnken设计及响应面分析确定主要影响因子的最佳浓度。结果表明:当葡萄糖、KH2PO4、玉米粉、黄豆粉、MnSO4、K2HPO4的最佳用量分别为0.72、1.073、.67、3.0、2.03、.0 g时,发酵培养基产生的芽孢数量达到最大值2.22×109cfu/mL,较优化前的1.81×109cfu/mL提高了22.7%。

Abstract:SAS system was used to optimize the medium components for Bacillus subtilis 504.Firstly,the prime factors affecting yield of Bacillus subtilis 504 were selected by means of Plackett-Burman design;Then,acentral composite design was used to optimize the prime factors and the results were shown inresponse surface polts.When the medium comprised glocose,KH2PO4,corn powder,soy bean powder,MnSO4,K2HPO4 yield of Bacillus subtilis 504 could reach 2.22 109cfu/mL,which was 22.7% higher than the control.
