| 摘 要： | 2b蛋白是黄瓜花叶病毒与植物寄主相互作用的一种重要多功能蛋白。用PCR方法扩增目的基因2b,经酶切、连接将其克隆至带有λcI基因的pBT载体上,构建与λcI基因融合的表达载体pBT-CMV 2b,转化大肠杆菌XL1-Blue MRF′报告菌株,用IPTG诱导表达黄瓜花叶病毒2b基因。Western blot分析结果表明,2b-λcI融合蛋白已成功表达,大小约为38 ku,与预期大小一致。pBT-CMV 2b与空质粒pTRG共转化XL1-Blue MRF′报告菌株,同时以pBT空质粒和pTRG-Gal11p共转化作为对照。结果显示,CMV 2b蛋白未产生自激活作用,从而为利用大肠杆菌双杂交系统筛选与2b蛋白相互作用的蛋白质研究奠定基础。
