绵羊生长激素基因第4外显子的多态性分析

摘    要:采用PCR-SSCP技术检测包括蒙古羊、小尾寒羊、德国美利奴羊以及德美寒杂交一代在内的绵羊生长激素(GH)基因第4外显子的多态性.结果表明:在1188~1502 bp片段上,德国美利奴羊及杂交后代中,AA型个体占多数,而小尾寒羊则以BB型个体为主.对这个片段的纯合型(AA,BB)分别进行克隆测序发现:1188~1502 bp片段上,AA型在第1406位发生了T→C的突变.实验结果证实绵羊生长激素基因第4外显子存在序列多态性.

Abstract:The polymorphism of ovine growth hormone gene in the 4th exon sequence in sheep including Mongolian,Small-tailed Han sheep,Germanmerino and F1 generation of the Small-tailed Han sheep and Germanmerino were detected by PCR-SSCP with a pair of primers.The results showed that in 1 188~1 502 bp most of the Germanmerino and hybrid generations presented as AA genotype,while most Small-tailed Han sheep presented as BB genotype.The amplified fragments of AA and BB were cloned and sequenced.The result showed that in 1 188-1 502 bp AA genotype had one substitution mutation at 1 406 site(T → C).The results confirmed that there were polymorphisms in the 4th exon region of GH gene.
