碱性溶液提取荞麦水溶性多糖的研究

摘    要:研究用碱溶液(0.3mol/LNaOH)提取养麦水溶性多糖,分别用料液比(1:30,1:40,1:50)和提取时间(1h,2h,3h),提取温度(70℃,80℃,90℃)3因素3水平正交法提取多糖,并研究了这3个因素对多糖得率的影响,方差分析表明温度对多糖提取影响较显著(P〈0.1),料液比和时间影响不显著。在本实验条件下,荞麦多糖的得率迭25.16%。同时用Sevag方法除蛋白,石油醚回流脱脂,淀粉酶,纤维素酶除去淀粉和纤维素,双氧水脱去色素对多糖进行了纯化。

Abstract:This study was on extracting polysaecharide from buckwheat by orthogonal design. Selected three factors (Solid- solvent ratio: 1 : 30,1 : 40,1 : 50; extracting temperature: 70℃,80℃ ,90℃ and extracting time: lh,2h, 3h). The results showed extracting temperature was significant (P〈0. 1) to extract buckwheat polysaccharide , then Solid-solvent ratio and extracting time were not significant(P〉0. 1). under this condition, polysaccharide extracting rate was 25.16%. Meanwhile, removed protein using Sevag method, removed oil using petroleum ether , removed amylum and cellulose using amylase and cellulase, removed pigment using H2O2 to purify polysaccharide.
