纳豆激酶植物高效表达载体的构建

摘    要:纳豆激酶来源于日本的传统食品—纳豆,是一种具有强烈溶栓作用的蛋白激酶,将编码纳豆激酶的基因克隆到含C aM V 35S控制下的双元表达载体pCAM B IA 1300中,PCR结果表明,纳豆激酶基因正向插入到载体中,再通过冻融法将其导入根癌农杆菌LBA 4404中,从而成功地构建了能在植物中高效表达的植物载体,为用植物作为生物反应器来获取纳豆激酶蛋白提供了一条新途径。

Abstract:Nattokinase,extracted from the Japanese food-natto, is found to be a strong fibrinolytic enzyme.The gene encoded Nattokinase was cloned into expressing vector-pCAMBIA1300 including CaMV35S promoter,PCR result indicated that NK gene was positively inserted,then the expression vector was transferred into Agrobacterium tumefaciens LBA4404 using the freeze-thaw method.Therefore,the vector,which could be efficiently expressed in plants,was successfully constructed,this can provide a new approach of gaining nattokinase protein by using plants as bioreactors.
