白蚁肠道木质素分解菌、染料脱色菌的分离鉴定及产酶条件优化

摘    要:分离具有木质素分解功能的菌株,并优化其产酶条件,同时观察分离菌株对染料的脱色情况,可为有效分解利用木质素提供必要的微生物资源。利用苯胺蓝平板法从黑胸散白蚁肠道中分离到2株具有木质素分解功能的菌株PY12、MX5,经16srRNA鉴定PY12、MX5分别为肠杆菌属的霍氏肠杆菌(Enterobacter hormaechei)、芽孢杆菌属的地衣芽孢杆菌(Bacillus licheniformis)。优化后菌株PY12LiP活力提高为278U/L,菌株MX5LiP活力提高为256U/L。并研究二者对6种染料的脱色效果观察,发现菌株PY12、MX5对6种染料都有不同程度的脱色作用,但对结晶紫脱色作用较弱。

Abstract:To provide necessary microbial resources for catalyzing lignin,the strains decomposing lignin were isolated and the fermentation conditions was optimized,the decolorizing effects of the isolated strains were investigated.The results showed that 2 strains,named PY12 and MX5,with decomposing lignin function were isolated using Azure B method from reticulitermes chinensis snyder gut.By 16 s rRNA identification,we found PY12 and MX5 belonged to Schizophyllum commune and Irpex lacteus,respectively.After optimizing the conditions of treatment to strains,the enzyme activities of PY12 and MX5 significantly increased to 278 U/L and 256 U/L,respectively.And PY12,MX5 showed some degree of decolorizing effects on six kinds of dyes,but they have a very weak decolorizing function to crystal violet.
