摘    要:鹅细小病毒(GPV)侵染引发机体酶或酶蛋白的变化,扰动平衡的代谢网络,调节多种代谢生理活动,其变化多样性和复杂性是研究病毒基因型与宿主遗传易感性互作病理学机制的有效标记物。通过鹅细小病毒YZ毒株(GPV-YZ)感染雏鹅,采用分光光度法分析脑、心、肝、肺、脾器官组织的蛋白酶、淀粉酶及转氨酶活性,聚丙烯酰胺凝胶电泳(PAGE)鉴定同工酶。结果显示,与对照组(CK)相比,受GPV感染雏鹅的各器官组织中,3种消化酶的活性分别增强1.6~6倍、1~2倍、2~3.6倍。淀粉酶同工酶存在4~7条不等的谱带变化,肝和脾新增1条慢区主带,而心脏缺少1个慢区谱带;转氨酶同工酶在慢区显示1条谱带,酶蛋白在慢中区谱带增多2~3条,而脑和脾脏的快区酶蛋白谱带减少1~2条。提示:慢区淀粉酶同工酶可能是GPV感染的敏感同工酶,酶蛋白/同工酶基因不同程度表达差异变化,直接或间接控制感染雏鹅的调节代谢生理机能,增强对入侵GPV的防御应激性能。

Abstract:Goose parvovirus (GPV) in fection results in the change of enzyme or zymoprotein of goslings, disturbing metabolic balance network and regulating many physiological activities and metabolism. The diversity and complexity of enzymes or zymoproteins were effective marker of pathological mechanism to investigate the interaction between virus genotype and host genetic susceptibility. Here, the protease, amylase and aminotransferase were respectively extracted from brain, heart, liver, lung, spleen organ of GPV-infected goslings and the control. The results showed that the activities of protease, amylase and aminotransferase in GPV-infected goslings was 1.6-6 times, 1-2 times, 2-3.6 times higher than those of the control group respectively. There were 4-7 bands of amylase isozymes in the organs of GPV-infected goslings. and appeared one new slow zone enzyme band of amylase isozyme in the liver and spleen of goslings infected GPV, but deleted one slow zone enzyme band of amylase isozyme in the heart of goslings infected GPV. There was one slow zone enzyme band of aminotransferase isozyme from goslings infected GPV and control group. There were increased 2-3 slow-medium zone zymoprotein band from goslings infected GPV, but deleted 1-2 fast zone zymoprotein band in the brain and spleen of goslings infected GPV. Those indicated that slow zone amylase isozyme might be the susceptible isozyme of GPV. The gene differential expression of zymoprotein/isozymes was controlled directly or indirectly affected the regulation of metabolic and physiological function on goslings infected GPV, and enhanced the defense and stress ability of GPV.
