摘    要:以标准株大肠杆菌K1 2 为材料对 3种细菌破壁方法 (SU法、ES法和ESU法 )进行了比较 ,并在此基础上测定了 17株病原菌的G +Cmol%含量。结果表明 :ESU法破壁提取的DNA含量最大 ,纯度最高 ,热变性曲线幅度最大 ,G +Cmol%含量符合文献值 ,17株病原菌鉴定至种 (11/ 17)或属 (6/ 11) ;在 3种破壁方法中 ,ESU法最有效 ,适用于水产动物病原菌的种属鉴定。

Abstract:Escherichia coli K_ 12 was used to pick out optimal method of breaking bacterial cellular wall,DNA qualities and G+Cmol% were compared among the three experimental ways,namely ES(lysozyme plus SDS), SU(SDS plus ultrasonic), ESU(lysozyme plus SDS and ultrasonic). ESU is the best method Judged by DNA purity and content,denaturation curve and G+Cmol%, 17 strains of bacteria were identified to species(11/17) or genus(6/17). Optimal method of breaking bacterial cellular wall was offered for G+Cmol% determination exactly and used for identification of aquatic pathogenic bacteria.
