摘    要:利用RAPD和ISSR分析30份果蔗种质的遗传背景,筛选出几个扩增条带清晰、多态程度较高、稳定可靠的引物(其中RAPD引物SS和ISSR引物WW可一次性准确地检测出黑皮果蔗、地方果蔗和自育杂交种间的区别),而且ISSR又比RAPD检测到更多的遗传多态性(二者的多态性条带比分别为61.35%和56.8%)。

Abstract:By means of RAPD and ISSR, the heredity backgrounds of 30 germplasmic resources of chewing cane were analyzed, some primers with clear amplifying, better polymorphic degree and stability were selected(Primer SS and WW could accurately examine the differences among badila, local chewing cane and hybridized varieties), and ISSR, compared with RAPD could examine more heredity polymorph(the ratios of the polymorphic bands were 61.35% and 56.8% respectively). The extraction of gene set DNA, PCR technique system, genetic diversities, classification, co-specific and hetero-named phenomenon etc. are discussed in this paper. In fine, this paper provides the theoretical basis for the identification and evaluation of the germplasmic resources, heredity classification, fingerprint diagram of chewing cane, which is helpful to the improvement of the study and use of the germplasmic resources of chewing cane.
