生防细菌A178的分子鉴定及抑菌机理的研究

摘    要:通过培养形态、生理生化特征以及16SrDNA的分子序列同源性分析,16SrDNA测序结果在GeneBank数据库进行同源性比较,确定A178菌株为枯草芽胞杆菌(Bacillus subtilis);采用平板对峙培养法,发现A178对棉花立枯病菌、棉花枯萎病菌和棉花黄萎病菌具有较强的抑菌作用,通过显微镜观察,发现拮抗细菌A178的主要拮抗机理是通过产生拮抗物质造成病原真菌菌丝体断裂、扭曲、畸形,抑制分生孢子的萌发,造成孢子畸形。

Abstract:Established the position of A178 strains based on the results of morphologic characteristics, physiological and biochemical properties and phylogenetic analysis of 16SrDNA sequence. Eventually A178 was identified as Bacillus subtilis. And A178 showed stronger antagonism against Rhizoctonia solani, Fusarium oxysporumf, sp. vasinfectum and Verticillium dahliaeon by plate duel culture. And the examination of antagonistic mechanism under light microscope showed that antagonistic substance of A178 appeared to inhibit pathogens by leading to mycelium rupture, distortion, abnormality, and inhibition bourgeon of conidiophore, and leading to abnormality of conidiophore.
