百合遗传多样性及亲缘关系的RAPD分析

摘    要: 对百合野生种及栽培品种共60份材料进行了RAPD分析, 22条随机引物共扩增出224条带,其中163条为多态性带, 多态率为72.77%。UPGMA分类结果将60份百合材料分为两大类群, 第1类群为铁炮与亚洲百合杂交系(L /A) 、野生种、亚洲系百合(A) 、喇叭百合杂交系(T) 及东方与喇叭百合杂交系(O /T) 的聚合, 这一类群在阈值0.668处又被分为两个亚类群; 第2类群为1 份野生种(L. speciosum ) 及铁炮系百合(L) 和东方系百合(O) 的聚合, 其中东方系百合在阈值0.746处又被分为两个亚类群。根据此分类结果, 进行了类群间亲缘关系及品种起源形成分析, 60份百合材料被划分为14个群组。经统计分析, 各群组之间的遗传相似系数与遗传距离的变化呈现相反的趋势。

Abstract:Sixty wild species and cultivars of L ilium ssp. were analyzed by RAPD. 22 random primers generated 224 bands in total, and 163 bands showed polymorphism, the average polymorphism rate was 72.77%. The dendrogram of the 60 lilymaterialswas generated byUPGMA (Unweighted Pair-group MethodArithmetic Average) method, and was divided into two groups. The first group included Longiflorum ×Asiatic hybrids, wild species, Asiatic hybrids, Trumpet hybrids, Oriental ×Trumpet hybrids, which was further divided into two subgroups. The second group was the clustering of L. speciosum , Longiflorum hybrids and Oriental hybrids. In addition, the Oriental hybrids were also divided into two small group s. Based on the results, the relationship between the groups, or subgroups, and the origin and derivation of the cultivars were analyzed. The 60 materialswere finally classified into 14 groups. It was found, by statistical analysis, that the genetic similarity between the groups presented an opposite trend with their genetic distance.
