微波预处理超声辅助酶解大豆秸秆条件优化

摘    要:为提高油料作物秸秆酶解效率,对大豆秸秆进行微波预处理,然后进行超声辅助酶解。经扫描电镜分析,大豆秸秆的致密结构经微波预处理后,得以明显破坏,可以更利于为纤维素酶水解。采用正交实验对微波预处理条件进行优化,结果表明微波预处理大豆秸秆最优条件为:微波辐射功率400W,辐射时间40min,辐射温度60℃。经微波预处理、超声辅助酶解条件优化后,水解7 h 酶解率达到11.06%,与常规条件酶解48 h 的酶解率(11.77%)基本相当,酶解效率显著增加。

Abstract:To improve the efficiency of enzymolysis, soybean straw was pretreated by using the microwave reactor and enzyme hydrolyzed by ultrasound-assisted method. By means of the scanning electron microscopy (SEM) analysis, the compact structure of soybean straw is evidently destructed after the microwave pretreatment, and was beneficial to hydrolyze by cellulase. Condition of microwave pretreatment is optimized by orthogonal experimental. As result, the optimal conditions for the microwave pretreatment of soybean straw are shown as follows: microwave irradiation power 400W, irradiation time 40 min, irradiation temperature 60 ℃. In optimized condition of the microwave pretreatment and ultrasound-assisted enzymolysis, the enzymatic hydrolysis yield of the straw is 11.06% at 7 h, which was almost equal as the yield (11.77%) of the conventional enzymolysis at 48 h. The enzyme efficiency increased significantly.
