大豆不同器官中的RNA的提取分析

摘    要:从植物组织中提取高质量的RNA是进行基因克隆和基因表达分析的基础.同种植物不同器官的组织由于组成成分的差别,其RNA的提取有不同的难点.本文分别采用异硫氰酸胍法、改进的异硫氰酸胍法提取了大豆的根、茎、叶、胚芽等不同器官的RNA,结果发现异硫氰酸胍法适合根和茎的RNA提取,改进的异硫氰酸胍法不仅适合根与茎的RNA提取,还适于胚芽RNA的提取.而两种方法对从叶中提取RNA则均不是很理想.

Abstract:Isolation of high-quality RNA is the base of studies on gene cloning and regulations on gene expression. Because of the difference in contents of components, the difficulties of isolating RNA differ from different plants or different organs of the same plant. We applied two methods of guanidine thiocyanat and an improved method by Wang Fang to find a suitable method for RNA isolation from embryo, leaf,stem and root of soybean. The results suggest that the method of guanidine thiocyanat fits for extracting RNA from soybean root and stem, and the improved method by Wang Fang fits for embryo as well as root and stem. Both of the two methods don't work very well for RNA isolation from soybean leafs.
