高活性纤维素酶的分离纯化

摘    要:为了纯化纤维素酶粗酶液,通过采用硫酸铵分级沉淀、Sephadex G-100凝胶柱层析、DEAF FF弱阴离子交换柱层析的方法,研究纤维素酶粗酶液分离纯化的方法。结果表明:采用硫酸铵分级沉淀、Sephadex G-100凝胶柱层析、DEAF FF弱阴离子交换柱层析的方法对内切葡聚糖苷酶组分的分离纯化有效。通过试验研究得到一种内切葡聚糖苷酶组分,其比活力提高了5.89倍,回收率为22%。

Abstract:To study the separation and purification of cellulase from crude enzyme,the method of ammonium sulfate fractionation,Sephadex G-100 gel filtration and DEAF FF weak anion exchange column chromatography method were used.The results showed that:the usage of ammonium sulfate fractionation,Sephadex G-100 gel column chromatography,DEAF FF weak anion exchange column chromatography was a efficient method to purify CMCase.The specific activity of CMCase increased 5.89 times purifed in this way,while the recovery rate was 22%.
