胡萝卜SOC1同源基因对光周期的响应及其功能

摘    要:基于先期抽薹敏感的‘松滋野生’和耐抽薹的‘Amsterdam’胡萝卜转录组测序结果,分析胡萝卜中SOC1同源基因对光周期的响应规律及其功能。胡萝卜中存在2个SOC1同源基因:DcSOC-1和DcSOC-2,具有完整的开放阅读框(ORF),分别编码217和211个氨基酸,均包含MADS-box和K-box结构域,C末端含有保守的SOC1 motif基序。系统进化树分析表明,DcSOC-1和DcSOC-2与猕猴桃Ac SOC1f和葡萄Vv SOC1的亲缘关系较近,属于SOC1/TM3亚家族。在长/短日照处理下,DcSOC-1在‘松滋野生’叶片中的相对表达量均显著高于‘Amsterdam’,且长日照能促进其表达,而DcSOC-2在‘松滋野生’和‘Amsterdam’中的表达水平较低。在春、秋两季生长过程中,DcSOC-1在‘松滋野生’叶片中的最高表达量分别达到522.4和401.7,而在‘Amsterdam’中均呈低水平表达,与先期抽薹率呈极显著正相关,说明DcSOC-1对胡萝卜的先期抽薹起着重要作用。

Abstract:Basis on the RNA-seq analysis,the relative expression and function of carrot SUPPRESSOR OF OVEREXPRESSION OF CONSTANTS 1(SOC1)homologues under photoperiod treatment were investigated with wild sensitive species‘Songzi’and cultivar‘Amsterdam’which was tolerant to premature bolting. There were two carrot SOC1 homologues:DcSOC1-1 and DcSOC1-2,which were annotated with complete open reading frames(ORF)encoding 217 and 211 amino acids respectively,which both containing MADS-box and K-box domain and a conserved SOC1 motif at the carboxy terminus. Phylogenetic analysis showed that DcSOC1-1 and DcSOC1-2 were closely related with kiwifruit AcSOC1f and grape VvSOC1,belonging to SOC1/TM3 subfamily. The expression of DcSOC1-1 in the leaves of‘Songzi’was significantly higher than that in‘Amsterdam’under short day and long day treatments,and long day can promote its expression,but the expression of DcSOC1-2 in‘Songzi’and‘Amsterdam’was low. During the growth of spring and autumn,the maximum expression values of DcSOC1-1 in‘Songzi’leaves were 522.4 and 401.7 respectively,but its expression was very low in levels of‘Amsterdam’. The correlation analysis revealed that the expression of DcSOC1-1 was significantly correlated with the ratio of premature bolting,which indicated DcSOC1-1 played an important role in carrot premature bolting.
