贝母愈伤组织的诱导及植株再生

摘    要:通过对三种野生贝母的离体培养,研究了贝母基因型、温光条件、激素配比对贝母愈伤及不定芽的影响.结果表明,在对贝母鳞茎外植体适当的低温暗处理后,再转入光下培养,可明显减轻外植体的褐化;不同基因型对贝母愈伤组织所诱导差异不大,进一步证明贝母愈伤组织的诱导,不依赖于基因型;贝母愈伤组织和不定芽的诱导需要不同的激素配比,诱导贝母愈伤组织的最佳培养基为(MS+6-BA 2.0 mg/L+NAA 0.5 mg/L),诱导不定芽的最佳培养基为(MS+6-BA 1.0 mg/L+2,4-D 0.1 mg/L),平均每个外植体均产生2~3个不定芽,且粗壮、移栽成活率高.建立了完整的不依赖于基因型的贝母植株再生体系.

Abstract:The genotypes, tempreture and light condition and hormonetype affecting induction of callus and shoots of Fritillariae were researched through culture in vitro of three wild Fritillariae. The result shows if bulbs were treated under the low tempreture,then cultured in the illumination, they can distinctly alleviate browning. Different genotypes have less diffrent induction on callus of Fritillariae. That further testifies the induction of callus doesn't depend on genotype, but the callus and shoos need different hormonetype. The medium with MS+6-BA 2.0mg/L+NAA 0.5mg/L is appropriate to induction of callus. Each average explant can produce 2~3 strong shoots on the medium with MS+6-BA 1.0mg/L+2, 4-D 0.1mg/L, moreever it has high transplanting rate. In the end we establish the full regeneration system without genotype of Fritillariae.
