青蒿组织培养的研究

摘    要:以青蒿茎段为外植体材料进行组织培养研究,结果表明:0.2%的HgCl2浸泡10 min对青蒿茎段材料灭菌有较好的效果;具腋芽的外植体在MS+6-BA(0.5~1.5)mg/L、MS+(0.01~0.05)mg/L TDZ、MS+1.0 mg/L 6-BA+0.2 mg/L IBA培养基上培养,腋芽都能萌发生长,在MS+(0.01~0.05)mg/L TDZ、MS+1.0 mg/L 6-BA+0.2 mg/L IBA培养基上培养,能诱导茎段产生大量不定芽;具有腋芽基部周围细胞的茎外植体在MS+0.5 mg/L 6-BA、MS+(0.01~0.05)mg/L TDZ培养基上培养,在其原腋芽处产生了大量丛芽。

Abstract:The stem of Artemisia annua was taken as explant to conduct tissue culture.The results showed that immersion of Artemisia annua stem in 0.2% HgCl2 for 10 min had better sterilization effect;culture of explant with axillary bud on media of MS+6-BA(0.5~1.5) mg/L,MS+TDZ(0.01~0.05) mg/L,MS+1.0 mg/L 6-BA + 0.2 mg/L IBA,the axillary bud can germinate and grow,on media of MS+TDZ(0.01~0.05) mg/L and MS+1.0 mg/L 6-BA+0.2 mg/L IBA can induce a large number of adventitious buds on stem;culture of explant with axillary bud basal cells on media of MS+6-BA 0.5 mg/L and MS+TDZ(0.01~0.05) mg/L,a large number of clumpy buds germinated from the position of original axillary bud.
