| 摘 要： | 以嗜热脂肪芽孢杆菌为初始菌种,经过诱变筛选,得到产异淀粉酶的突变菌株UM761,酶活在原始菌种产酶水平的基础上提高了2~3倍。对UM761的产酶条件进行了优化,结果表明:最适产酶菌株培养条件为以1%葡萄糖作为碳源,牛肉膏、蛋白胨和酵母粉的混合物作为氮源,温度44~46℃,培养时间48~64 h,培养基初始pH值6.2~6.5。金属离子Co2+、Mg2+、Ca2+对异淀粉酶有较强的激活作用,而Fe3+、Fe2+对酶则有较强的抑制作用。

| Abstract: | A mutated strain UM761,which can produce isoamylase,was obtained from induced screening of Bacillus stearothermophilus.The activity of isoamylase produced by UM761 was enhanced by 2~3 times than the activity of isoamylase produced by Bacillus stearothermophilus.Meanwhile,the conditions for UM761 producing isoamylase were optimized,and the results showed as follows: taking 1% glucose as carbon source and the mixture of beef extract,peptone and yeast powder as nitrogen source;the optimum cultivation temperature and time were 44~46℃ and 48~64 h,respectively;the incipient pH value for culture medium ranged from 6.2 to 6.5.Co2+,Mg2+ and Ca2+ have strong activation effect on activity of isoamylase,but Fe3+ and Fe2+ have inhibitory effect on that. |
