摘    要:应用RT-PCR方法,以1株H5N1亚型禽流感病毒RNA为模板,扩增了NA全基因cDNA。将NA cDNA克隆于pMD18-T栽体,并对其进行了测序。测序结果表明:所克隆的NA基因全长为1416bp,编码区为1410个核苷酸,编码469个氨基酸。将其序列与5株H5N1亚型禽流感病毒NA基因序列进行比较,其核苷酸和氨基酸同源性均为96.6%~99.1%。NA的克隆和分子进化分析揭示了各毒株间的亲源关系。

Abstract:In this study, full-length neuraminidase(NA) gene was amplified from an avian influenza virus isolate(H5N1) by RT - PCR. The cDNA of NA gene was cloned into pMD18-T vector and then was se-quenced. The cloned 1 416 bp fragment covered the whole NA gene including the completely open reading frame which encoded 469 amino acids. The results of comparative sequence analysis indicated that the nucleotide sequence of NA gene of this isolate has 96.6 % ~ 99.1 % homology with those of other three H5 N, 1 isolates. The putative amino acid sequence has 96. 6 % ~ 99.1 % homology with those of other five H5 N1 isolates. This study revealed the relationship among these isolates.
