温度对甘蓝型油菜花粉胚状体的诱导及其类型的影响

摘    要:为了提高甘蓝型油菜(Brassica napus L.)花粉胚状体的诱导频率,进行了花药接种前的低温处理和接种后的高温处理,试验采用了五种不同温度的分段培养,对比了变温处理的效应。结果表明:不同基因型油菜花药培养,对变温处理的反应有差异;低、高温处理花药,对胚状体的类型有明显的影响;根据胚状体的形状和大小不同可分为五类,它们的再生植株能力不同。变温处理可以提高甘蓝型油菜花粉胚状体诱导频率和正常胚比率。

Abstract:Anthers were treated under low temperature before inoculation and high temperature after inoculation for enhancing induction frequency of pollen embryoid in Brassica napus L.. In this experiment five different temperature treatments were used. The result showed that anthers from different genotypes had different reponses by change of temperature, and induction freaueney of pollen embryoid from anthers treated under low and high temperature was higher than that of the untreated fresh anthers.It was also observed that the changing temperature treatment had obvious influence to the embryoid types and that based on the different morphologies and sizes the pollen embryoids could be divided into five types which had different regeneration frequencies. It is an effective way that changing temperature treatment enhanced induction frequency of pollen embryoid and normal embryoid ratioes.
